丝胶通过上调 Nrf2/HO-1 和 HSP90 的表达保护玻璃化冷冻小鼠卵巢组织

摘要

目的:探究丝胶是否通过调控HSP90和Nrf2/HO-1通路来保护玻璃化冷冻卵巢组织。方法:取30只小鼠卵巢,随机分新鲜组、对照组、0.5%丝胶组、1%丝胶组和0.1mM褪黑素组(M LT),除新鲜组外,其他四组均进行玻璃化冷冻。进行苏木素-伊红染色(HE)及免疫组织化学染色(IHC)。玻璃化冷冻的四组进行蛋白免疫印迹(WB)和实时荧光定量PCR实验。结果:丝胶组可改善卵泡细胞形态,维持较高的形态正常率。IHC显示丝胶组增加Bcl-2的表达,降低Bax和Caspase-3的表达。WB和PCR结果显示丝胶组可有效提高Nrf2/HO-1、HSP90蛋白和mRNA的水平。以上结果1%丝胶效果最佳,与M LT无显著差异。结论:丝胶可提高玻璃化冷冻卵巢的抗氧化能力,降低细胞凋亡,维持卵巢的正常结构,其作用可能与上调Nrf2/HO-1通路及HSP90的表达相关。
