肿瘤相关成纤维细胞的促癌机制及治疗药物研究进展

摘要

肿瘤相关成纤维细胞(Cancer Associated Fibroblasts,CAFs)是大多数实体肿瘤微环境的重要组成部分,通过支持肿瘤细胞生长、细胞外基质重塑、促进血管生成和介导炎症反应等途径促进癌症进展。CAFs还可以通过多种机制促进肿瘤的免疫逃逸,包括多种细胞因子和趋化因子的分泌以及介导先天和适应性免疫细胞募集和功能分化。目前,针对肿瘤细胞的化疗存在一定局限性,其不能根除肿瘤细胞并使其继续增殖及产生转移灶。同时存在一些同时靶向肿瘤细胞及CAFs或者单独靶向CAFs的治疗方式。论文重点介绍了CAFs促癌机制的最新研究进展,以及针对CAFs的治疗方法,以期将临床前及临床发现整合到抑制CAFs的肿瘤支持功能的新治疗策略中。
