激活素A抑制人骨髓瘤RPMI-8226细胞增殖的机制研究

摘要

激活素 A 属于 TGF-β 超家族成员,具有抑制小鼠骨髓瘤细胞增殖、促进凋亡的作用,但对人骨髓瘤细胞的作用尚不清楚。本研究利用人骨髓瘤细胞系 RPMI-8226 细胞,采用 Cell Counting Kit-8 法检测激细胞活力,流式细胞术检测凋亡以及 PCR技术检测 mRNA 表达。实验结果发现,激活素 A 能够抑制 RPMI-8226 细胞活力,诱导 RPMI-8226 细胞凋亡,并且能够促进 ActRIIA 和 Smad3 的表达,提示激活素 A 通过作用于 ActRIIA-Smad3 抑制 RPMI-8226 细胞增殖。
