结直肠腺瘤相关的长链非编码RNA差异表达谱建立及应用

摘要

目的:筛选结直肠腺瘤组织样本中具有差异表达的长链非编码RNA(long non-codingRNA,LncRNA),预测其靶基因,探究靶基因的生物学功能及富集的信号通路。方法:收集临床结直肠手术中腺瘤新鲜样本,高通量测序技术构建LncRNA差异表达谱,生物信息学技术筛选差异表达的LncRNA并进行靶基因预测,GO与KEGG富集分析分别预测靶基因的生物学功能及富集的信号通路。结果:与正常组织样本相比,结直肠腺瘤组织样本中筛选出219个显著差异表达LncRNA(119个上调,99个下调)。功能性分析表明:与结直肠腺瘤相关的GO条目1242条;KEGG富集的信号通路32条。结论:结直肠腺瘤样本中差异表达的LncRNA可能通过靶向调控其靶基因参与调控多种生物学功能及结直肠腺瘤相关的信号通路,在结直肠腺瘤形成过程中可能发挥重要作用。
